ABSTRACT
Streptavidin is the pragmatic protein of choice for better understanding both intramolecular and intermolecular interactions and for versatile use in biotechnological applications. Here we present two high-resolution apo-streptavidin structures at both ambient temperature (Apo-SFX; 1.7 Å; LCLS) and cryogenic temperatures (Apo-Cryo; 1.1 Å; SSRL). In this study, we present the new cooperative allosteric perspective, in which streptavidin binds to the substrate through water molecules characterized by a polar interaction network and mimics the substrate biotin.
