Abstract:
Streptomycin, an aminoglycoside antibiotic, binds close to the codon recognition site, inducing translational errors by disrupting the decoding. It interacts with 16SrRNA and ribosomal S12 protein, binding to the 30S ribosomal subunit. In this study, we aim to explore the effect of streptomycin binding to the decoding center and its interaction with ribosomal protein S12 of the X-ray crystal structures of the 30S ribosomal subunit in order to reveal the underpinning mechanism of streptomycin action at ambient temperature. Streptomycin-bound 30S ribosomal subunit causes a vital distortion over 16S ribosomal RNA, including A1492 and A1493 bases that are closely related to codon recognition. Additionally, revealed a correlation between shoulder movement up to 1 Å pairwise distance and h44–h45 engagement compared to the apo-30S ribosomal subunit, providing some insight into how streptomycin distorts the decoding center. The determination of this time-resolved SFX data at XFEL provided opportunities to obtain further structural insight into the streptomycin-30S ribosomal subunit interaction and function, providing to development of a robust approach against streptomycin resistance.
